The best HIC column for each separation:
BioPro HIC HT and BioPro HIC BF
In order to address separation requirements of different compounds, YMC offers two different HIC columns, BioPro HIC HT and BioPro HIC BF. They both have in common their non-porous polymethacrylate particle, while their properties and chromatographic behaviours are quite different.
In this application note the retention behaviour of BioPro HIC HT and BioPro HIC BF columns is compared by separating two commercial monoclonal antibodies from each other. As BioPro HIC HT is designed for the analysis of biopharmaceuticals, it is the first choice for ADCs or MAbs. It provides high resolution with very fast runs and therefore high throughput. BioPro HIC BF columns show a stronger retention due to their higher hydrophobicity. They can therefore be used for the separation of low hydrophobic proteins or especially for the analysis of oxidised MAbs.

Characterisation of monoclonal antibodies
Do you analyse MAbs using HIC or determine the drug-to-antibody ratio of ADCs?
Are you interested to learn how to couple HIC and MS?
This product information shows how to use HIC and MS online. In addition, you can find out how YMC’s innovative column BioPro HIC BF helps to overcome the high salt concentrations.

Native online SCX-MS analysis of monoclonal antibodies
Are you looking to avoid the usual two-step process of combining cation exchange chromatography (CEX) with mass detection (MS), where the specific charge variant peak of interest has to be isolated prior to characterisation via mass spectrometry?
Would you like to couple CEX to MS directly to save time and exclude possible artefacts of the long isolation process?
Learn more about high resolution SCX coupled directly to an ultrasensitive online native MS. The combination of MS compatible buffer and YMC’s non-porous BioPro IEX SF column allows the characterisation of the NISTmAb (and five in-house MAbs with different pI). The mobile phase composition and gradient were optimised to achieve an efficient separation of charge variants.
