Generic Analyses of Different Monoclonal Antibodies with RP-UHPLC

Due to their molecular weight of about 150 kDa, intact antibodies are usually analysed by IEX, SEC or HIC. In addition, RP methods have become an easy tool which is compatible with mass spectrometry (MS). Successful analysis in RP mode for MAbs is enhanced by employing a widepore, temperature-stable (>60°C) stationary phase, such as YMC-Triart Bio C4.

This application note shows how to achieve robust chromatographic results for 5 commercially available MAbs: Trastuzumab (Herceptin®), Rituximab (MabThera®), Adalimumab (Humira®), Bevacizumab (Avastin®), and Nist mAB. This generic analysis is carried out in less than 7 min using 1.9 μm particles for UHPLC and an elevated temperature of 80 °C resulting in higher sensitivity and sharper peaks for all the MAbs analysed.

YMC: Your Reliable Partner for Preparative Stationary Phases!

Reliability and efficiency: Unique advantages of YMC preparative media

YMC stationary phases are used in many preparative industrial purifications. They are selected because of their robustness, reproducibility and capacity. The longevity of our base material and the resulting cost effectiveness are also key factors in their selection. With over 35 years experience, YMC has also become a competent partner for regulatory support. As a result, YMC phases are employed worldwide in long-term processes where a reliable supply is guaranteed. This applies in equal measures to both the kg- as well as the multi-ton scale.
