How to determine the purity of a crude oligonucleotide

A practical example for qualification and quantification of a complex oligonucleotide sample by IP-RP UHPLC prior to further purification has been produced. This very comprehensive approach had been published by Fioretti et al.

YMC-Triart C18 UHPLC in IP-RP mode.

Essential guideline for robust HIC methods for proteins and mAbs

A recently published protocol paper summarises how to set up robust HIC methods. The paper focuses on the analysis of intact proteins and mAbs. Prof. Eeltink’s group (University of Brussels/VUB) present helpful and detailed aspects especially for:

  • How to set-up and maintain the system to achieve robust performance
  • The effect of sample-preparation conditions on sample breakthroughs
  • Run-to-run repeatability for 5 different columns obtained from 3 different column lots

The samples analysed are a mixture of intact proteins or monoclonal antibodies. The column of choice is YMC’s BioPro HIC HT column.

Analysis of denaturated and non-denaturated siRNA via IP-RP and AEX

siRNAs are a powerful tool for genetic analysis and treatment of metabolic diseases. siRNAs can be analysed as duplex by HPLC under non-denaturing conditions to control duplex formation and the amount of excess single strands. In addition, non-hybridized single stranded RNA can be studied under denaturing conditions. In both cases, IP-RP and AEX are the most commonly used methods.

In this Application Note the analysis of a siRNA duplex under denaturing and non-denaturing conditions is shown. A bioinert YMC-Accura Triart Bio C18 UHPLC column is used in IP-RP mode, while a BioPro IEX QF column is used in AEX mode.
